Flow cytometry is based on the study of optical parameters derived from the interaction of an excitation light source (laser) with particles (cells, subcellular fractions). In this way, structural information (size, complexity) can be collected and, from the fluorescent labeling of different epitopes, multiparametric information can be obtained from each individual cell of the general population. Based on these properties, cytometry techniques make it possible to analyze and classify a considerable number of cells, with relative ease and speed, with objectivity and high sensitivity, to obtain qualitative and quantitative measurements of complex subcellular populations. Resources and additional information The High Technology Unit (UAT) is a VHIR’s preclinical core-facility that provides researchers with state-of-the-art equipment and specialized technical advice, offering different services within its five Platforms (Genomics, Cytometry, Molecular Diagnosis, Metabolomics and Microscopy) This platform aims to offer the scientific community services and protocols related to functional genomics, mainly dedicated to the profile of gene expression using microarrays: This platform is complemented by the services of our genomics platform, both for quantitative analysis and qualitative of nucleic acids as for the validation of the results obtained by qPCR.  

Services

Cell analysis by flow cytometry

• FACS (Fluorescence-Activated Cell Sorting), for the isolation of specific cells or populations.

The UAT offers advice on experimental design and data analysis, technical support and general training in cytometry, including the handling of self-service cytometers. Tailor-made experiments (including labeling) can be carried out on request, after consultation with Unit staff.

APLICATIONS

• Immunophenotyping based on the labeling of surface and intracellular markers

• Studies derived from the labeling of nucleic acids (ploidy, cell cycle)

• Functional analysis (enzymatic activity, cell integrity, membrane permeability, calcium fluxes)

• Identification and isolation of cell populations or particles (cell classification)

Equipment

  • Facs Aria Digital Cell Sorter (BD): 4 lasers to detect up to 12 fluorochromes simultaneously.
  • Two FacsCalibur (BD) cell analyzers: 2 lasers for simultaneous detection of up to 4 fluorochromes.
  • Fortessa LSR (BD) cell analyzer: 4 lasers and parallel detection of up to 16 fluorochromes. Acquisition of high performance samples (96 and 384 well plates).
  • Data analysis software: FCS Express (online licenses) and FlowJo (installed on a UAT computer).
